摘要: 为研究紫竹Phyllostachys nigra多酚氧化酶基因(PPO)表达与紫竹组培苗褐化之间的关系,采用同源克隆方法获得紫竹PPO基因的cDNA与DNA片段,将它命名为PnPPO。随后,研究了该基因在不同褐化程度紫竹组培苗中的表达情况。序列分析显示该基因所编码蛋白为酪氨酸酶(tyrosinase),其序列与小麦Triticum aestivum和水稻Oryza sativa中PPO基因同源性很高,是PPO的同源序列。RT-PCR分析显示,在褐化严重的组培苗中PnPPO的表达量也较高。这说明PnPPO基因的表达与紫竹组培苗褐化存在紧密的联系。图4参15Abstract: To study the relationship between polyphenol oxidase (PPO) and tissue browning in Phyllostachys nigra,homologous cloning was used to obtain a partial sequence of the PPO gene in Phyllostachys nigra (named PnPPO). The gene structure and the expression pattern in tissue cultured seedlings with different degrees of browning were checked using reverse transcription Polymerase Chain Reaction(RT-PCR). The sequence revealed a homolog of the PPO gene in Oryza sativa and Triticum aestivum with the amino acid belonging to tyrosinase. Also,RT-PCR results showed that with high levels of browning,PnPPO had strong expression. Thus,the expression of PnPPO was related to the degree of browning.[Ch,4 fig. 15 ref.]
